A sample preparation workshop was convened at the 20th International Radiocarbon conference, with about 30–40 attendees. The term “sample preparation” was not further specified to allow participants to address the laboratory issues most important to them. Given the short time available, the focus quickly narrowed to details of graphite preparation for accelerator mass spectrometry (AMS), with particular interest in procedures for small samples, and hence we have changed the title to reflect the focus. Here, we summarize the workshop discussion, adding additional background information and references to publications. Several aspects of graphite preparation are included: catalyst types and ratio of catalyst to carbon; optimal reaction temperatures for various sample sizes; methods for water removal; types of pressure transducers; preconditioning of catalyst; and problems with blank values in very small samples. The pros and cons of sample dilution were also discussed.

We are developing practical methodologies to characterize pool sizes and residence times for fractions of soil organic matter (SOM) using radiocarbon, with a particular focus on SOM in New Zealand pasture soils that responds to global change on decadal timescales. As single mean residence times for the entire SOM pool can be misleading or uninterpretable, we focus on the use of samples collected about 7 and 40 yr after the bomb14C spike to separate SOM into at least 2 pools. These results from a box model methodology yield sensible estimates of the proportion of “passive” SOM, and the residence time of the dominant pool with approximately decadal residence times. These results are supported by chemical analysis. Approximately 45-yr residence times of light-fraction SOM in a relatively infertile soil contrast with ∼16-yr residence times in a more fertile soil, and correspond to large differences in the proportion of lignin- and polysaccharide-derived SOM in these soils measured using pyrolysis-GC/MS. To achieve greater detail and assess the degree to which “active” SOM with annual turnover rates may bias results from the simple model, we use density as a means of isolating SOM with different degrees of mineral association. Initial results from grazed pasture soils sampled in 2003–4 emphasize that isolating non-mineral-associated light fractions can improve understanding, but may be less important than identifying fractions associated with unique mineralogy. In this soil, a fraction with density ≥2.55 g/mL shows much larger proportions of passive SOM than other fractions.
An AMS 14C Pollen-Dated Sediment and Pollen Sequence from the Late Holocene, Southern Coastal Hawke'S Bay, New Zealand

Hawke's Bay is a region of New Zealand where earliest settlement of indigenous people may have occurred. A sedimentological and palynological study of lake sediments from a small catchment was undertaken to reconstruct erosion, vegetation, and fire histories to determine human environmental impact, and thus add to knowledge of the timing of initial settlement of New Zealand. Precise dating was an essential facet of the research because of the short time span of human occupation in New Zealand. A chronology is proposed based on accelerator mass spectrometry (AMS) radiocarbon dating of palynomorph concentrates. Known-age tephras were used as a check on the validity of the 14C ages obtained using this technique, which is being developed at Rafter Radiocarbon Laboratory. Two episodes of sustained erosion occurred between about 1500 and 1050 BC with a period of ~50 yr at about 1300 BC when no erosion occurred. Five episodes of erosion of very short duration occurred at about 625 BC, 450 BC, 100 BC, AD 950, and AD 1400. Erosion probably resulted from landslides induced by earthquakes or severe storms, with the exception of the last event which coincides with local burning and is probably a consequence of this. A conifer/broadleaved forest surrounded the lake until soon after AD 1075–1300, when a dramatic decline in pollen of forest plants and an increase in charcoal occurred. Forest was replaced by fire-induced scrub, interpreted as a result of anthropogenic burning by prehistoric Polynesians. A further decline in woody vegetation occurred when European-introduced plants appear in the pollen record and extensive pasture was established.
Anomalous Radiocarbon Dates from Easter Island

The largest volcanic crater on Easter Island in the South Pacific contains a lake 1 km in diameter with large floating mats of vegetation, mainly Scirpus californicus. A core taken through a mat near the center produced anomalous dates, with older dates above younger ones. The possibility that the mat had become inverted was considered, but palynological evidence refutes this idea because it shows a progressive upward decline of forest pollen, which is well known from other swamp cores on the island. A new series of radiocarbon dates made directly on pollen concentrates was obtained. These dates also produced inconsistencies, particularly when pollen concentrate ages were compared with 14C ages on plant fragments from the same depth. This series of 14C ages seems to indicate that both old and young organic components in the sediment are deposited contiguously and that the depositional history of these cores is more complex than previously known. Previous age determinations on bulk sediments from Easter Island, which also show anomalous dates, may be too simplistic. This paper provides a warning to other researchers dating sediments from Easter Island. We suggest that sample selection and dating procedures be carefully considered for these sediments.

A simple method for preparing pollen concentrates for 14C AMS dating is applied to organic and inorganic deposits from a peat bog in south Westland, New Zealand, from which preliminary AMS dating indicated age inversions and severe younger carbon contamination problems. The AMS ages of the pollen concentrates provided consistently older age estimates for each sample than ages derived from their respective organic residue or combined pollen and organic residue fractions. It is likely that the younger age estimates of the organic residue fractions result from the incorporation of younger plant material into the sample and possible contamination from younger humic acids percolating through the site.

A series of 14C determinations have been obtained on hair samples principally from Holocene contexts that have been variously pretreated to examine different means of removing potential contamination. SEM photomicrographs have documented hair surfaces before and after different pretreatments. Amino-acid composition, C/N ratios and δ13C values have been obtained to biochemically characterize these samples and provide baseline data for future comparisons with less well-preserved samples. Our data support the view that appropriately pretreated hair samples can provide accurate 14C age determinations.

Procedures and equipment used in the University of Wisconsin laboratory have been described previously (Bender, Bryson & Baerreis 1965; Steventon & Kutzbach 1986). Except as otherwise indicated, wood, charcoal and peat samples are pretreated with dilute NaOH-Na4P2O7 and dilute HCl before conversion to counting gas methane; when noted, marls and lake cores are treated with acid only.

The East-Anglian CTC Trainees' Day was held at Friern Hospital, London, on 14 December 1989. The programme comprised a morning session with two speakers and lunch sponsored by Duphar, followed by an open forum on CTC training issues and an address by the final speaker. Approximately 30 trainees attended, mostly SHOs and registrars. Professor A. Wakeling, chairman of the East-Anglian Division of the Royal College, was present and took the opportunity to meet trainees from the region.
